We demonstrate that engraftment of microglia-like cells following microglial depletion exacerbated EAE in Cx3cr1CreER/+Rosa26DTA/+ female mice as assessed by clinical symptoms and the expression of CNS inflammatory factors, but these findings were not evident in male mice. Higher major histocompatibility complex class II expression and cytokine production in the female CNS contributed to the sex-dependent EAE severity in mice following engraftment of microglia-like cells. An underestimated yet marked sex-dependent microglial activation pattern may exist in the injured CNS during EAE.It is well-known that multiple functional miRNAs are found in mammals' ovaries, which are linked not only to ovarian development, but also to maturation and apoptosis. However, there is still a lack of knowledge regarding the role of miRNAs in the hen ovary. In the present study, we analyzed the miRNA sequencing libraries of ovaries at the four different developmental stages of hens (15, 20, 30, and 68 W) and a total of 677 known miRNAs and 61 novel miRNAs were identified. In total, 209 of them were differently expressed miRNAs (DE miRNAs) obtained from comparisons of the four stages, including 84 upregulated and 125 downregulated DE miRNAs. Furthermore, the five key DE miRNAs gga-miR-2954, gga-miR-6634-5p, gga-miR-449b-5p, gga-miR-449c-3p, and gga-miR449c-5p were screened using an analysis of the miRNA-mRNA interaction network and functional enrichment annotated in seven significantly enriched pathways, such as endocytosis, lysine degradation, the biosynthesis of amino acids, and the MAPK signaling pathway, which may primarily participate in cell differentiation and proliferation, steroid hormone biosynthesis, and angiogenesis by targeting the related genes. For instance, gga-miR-449 family members were predicted to target 15 genes, including TGFB1, TPM1, TPM3, and CAMKB2, which were reported to regulate follicular growth, selection, and the ovulatory cycle. Taken together, our results illustrate the ovarian miRNA profiles of the four classic developmental stages of hens and highlight the significant role of miRNAs in ovarian development and functions. However, in-depth research needs to be carried out to validate the potential functional miRNAs found in this study.Accurate analysis using a simple and rapid procedure is always the most important pursuit of analytical chemists. In this study, a new sample preparation procedure, namely the shaker cup (SH) method, was designed and compared with two sample preparation procedures, commonly used in the laboratory, from three aspects homogeneity of the sample-flux mixture, potential for sample contamination, and sample preparation time. For the three methods, a set of 54 certified reference materials (CRMs) was used to establish the calibration curves, while another set of 19 CRMs was measured to validate the results. In the calibration procedures, the matrix effects were corrected using the theoretical alpha coefficient method combined with the experimental coefficient method. The data of the major oxides (SiO2, TiO2, Al2O3, TFe2O3, MnO, MgO, CaO, Na2O, K2O, and P2O5) and minor elements (Cr, Cu, Ba, Ni, Sr, V, Zr, and Zn) obtained by wavelength dispersive X-ray fluorescence spectroscopy (WD-XRF) were compared using two derivative equations based on the findings by Laurence Whitty-Léveillé. Understanding the ecological interactions between Varroa and honeybees is fundamental for reducing mite impact in apiaries. This work assesses bee colonies with various Varroa infestation levels in apiaries to determine (1) the relationship between multi-infested brood cells and brood infestation level, (2) the damage caused by Varroa to parasitized honey bee pupae, and (3) mite reproduction rate at various infestation levels. Data were collected from 19 worker brood combs, each from a different colony, ranging from 160 to 1725 (mean = 706) sealed cells per comb. Mite distribution was aggregated, ranging from about 2% to 74% infested cells per comb. The percentage of cells invaded by one, two, three, four, or more than four foundress mites, as a function of infestation level, was estimated by five highly significant (p less then 0.0001) second-degree polynomial regression equations. The correction factors found could increase the precision of prediction models.
