Two cubs were born naturally after 90 days of gestation, representing the first cheetah births resulting from transfer of embryos produced in vitro.Globally, one of the biggest problems with the increase in the elderly population is dementia. However, dementia still has no fundamental cure. Therefore, it is important to predict and prevent dementia early. For early prediction of dementia, it is crucial to find dementia risk factors that increase a person's risk of developing dementia. In this paper, the subject of dementia risk factor analysis and discovery studies were limited to gender, because it is assumed that the difference in the prevalence of dementia in men and women will lead to differences in the risk factors for dementia among men and women. This study analyzed the Korean National Health Information System-Senior Cohort using machine-learning techniques. By using the machine-learning technique, it was possible to reveal a very small causal relationship between data that are ignored using existing statistical techniques. By using the senior cohort, it was possible to analyze 6000 data that matched the experimental conditions out of 558,147 sample subjects over 14 years. In order to analyze the difference in dementia risk factors between men and women, three machine-learning-based dementia risk factor analysis models were constructed and compared. As a result of the experiment, it was found that the risk factors for dementia in men and women are different. In addition, not only did the results include most of the known dementia risk factors, previously unknown candidates for dementia risk factors were also identified. We hope that our research will be helpful in finding new dementia risk factors.Although epidemiological studies have shown a relationship between periodontal disease and pancreatic cancer, the molecular mechanisms involved remain unclear. In this study, the effects of systemic administration of Porphyromonas gingivalis lipopolysaccharide (PG-LPS) on gene expression were comprehensively explored in mouse pancreas that did not demonstrate any signs of inflammation. PG-LPS was prepared in physiological saline and intraperitoneally administered to male mice at a concentration of 5 mg/kg every 3 days for 1 month. After extracting total RNA from the excised mice pancreas, a comprehensive DNA microarray analysis of gene expression was performed. Tissue specimens were also subjected to hematoxylin-eosin staining and immunohistochemistry using anti-regenerating islet-derived 3A and G (Reg3A/G) antibody. ImageJ software was used to quantify the area of Reg3A/G positive cells in pancreatic islets by binarizing image date followed by area extraction. The results were compared using Mann-Whitney U test. Data are presented as mean ± standard deviation (SD) with p less then 0.05 considered as significant. Reg3G, a gene related to pancreatic cancer, was one of the 10 genes with the highest levels of expression in the pancreas stimulated with PG-LPS. The comprehensive analysis revealed a 73-fold increase in Reg3G expression level in the PG-LPS group when compared with the control group; in addition, the expression level of Reg3A was increased by 11-fold in the PG-LPS group. Image analysis showed that the ratio of Reg3A/G positive cells was higher in the PG-LPS group than the control. Immunostaining showed the presence of Reg3A/G-positive cells in the alpha-cell equivalent areas around the islets of Langerhans in the PG-LPS group. These results support the notion that periodontal disease may be a risk factor for pancreatic cancer.Chloroethylagelastatin A (CEAA) is an analogue of agelastatin A (AA), a natural alkaloid derived from a marine sponge. It is under development for therapeutic use against brain tumors as it has excellent central nervous system (CNS) penetration and pre-clinical therapeutic activity against brain tumors. Recently, AA was shown to inhibit protein synthesis by binding to the ribosomal A-site. In this study, we developed a novel virtual screening platform to perform a comprehensive screening of various AA analogues showing that AA analogues with proven therapeutic activity including CEAA have significant ribosomal binding capacity whereas therapeutically inactive analogues show poor ribosomal binding and revealing structural fingerprint features essential for drug-ribosome interactions. In particular, CEAA was found to have greater ribosomal binding capacity than AA. Biological tests showed that CEAA binds the ribosome and contributes to protein synthesis inhibition. Our findings suggest that CEAA may possess ribosomal inhibitor activity and that our virtual screening platform may be a useful tool in discovery and development of novel ribosomal inhibitors.Myeloid-derived suppressor cells (MDSCs) are major immunosuppressive cells in the tumor microenvironment (TME). #link# During the differentiation and development of MDSCs from myeloid progenitor cells, their functions are also affected by a series of regulatory factors in the TME, such as metabolic reprogramming, epigenetic modification, and cell signaling pathways. Additionally, there is a crosstalk between these regulatory factors. This review mainly introduces the metabolism (especially glucose metabolism) and significant epigenetic modification of MDSCs in the TME, and briefly introduces the connections between metabolism and epigenetic modification in MDSCs, in order to determine the further impact on the immunosuppressive effect of MDSCs, so as to serve as a more effective target for tumor therapy.The superior fracture toughness of zirconia is closely correlated with stress-induced martensitic phase transformation around a crack tip. In this study, a modified phase field (PF) model coupling phase transformation and fracture is proposed to study the fracture behavior and toughening effect of tetragonal zirconia polycrystal (TZP). The stress-induced tetragonal to monoclinic (t-m) phase transformation around a static or propagating crack is characterized with PF simulations. It is shown that the finite size and shape of the transformation zone under different loads and ambient temperatures can be well predicted with the proposed PF model. The phase transformation may decrease the stress level around the crack tip, which implies the toughening effect. After Sacituzumab govitecan concentration , crack propagation in TZP is studied. As the stress field is perturbed by the phase transformation patterns, the crack may experience deflection and branching in the propagation process. It is found that the toughness of the grain boundaries (GBs) has important influences on the crack propagation mode.
