CAR-T cell therapy, as a novel immunotherapy approach, has indicated successful results in the treatment of hematological malignancies; however, distinct results have been achieved regarding solid tumors. Tumor immunosuppressive microenvironment has been identified as the most critical barrier in CAR-T cell therapy of solid tumors. Developing novel strategies to augment the safety and efficacy of CAR-T cells could be useful to overcome the solid tumor hurdles. Similar to other cancer treatments, CAR-T cell therapy can cause some side effects, which can disturb the healthy tissues. In the current review, we will discuss the practical breakthroughs in CAR-T cell therapy using the multi-targeted and programmable CARs instead of conventional types. These superior types of CAR-T cells have been developed to increase the function and safety of T cells in a controllable manner, which would diminish the incidence of relevant side effects. Moreover, we will describe the capability of these powerful CARs in targeting multiple tumor antigens, redirecting the CAR-T cells to specific target cells, incrementing the safety of CARs, and other advantages that lead to promising outcomes in cancer CAR-T cell therapy.Calciphylaxis is a deadly, painful disease with a 1-year mortality of up to 50%. The disease is commonly associated with patients with end-stage kidney disease (ESKD), but it can manifest in non-uremic patients as well. In patients who are undergoing dialysis, the incidence of calciphylaxis can range from 0.04% to 4%. The progressive arterial calcification seen in calciphylaxis can affect multiple body organs, including the skin, brain, lungs, and muscle. In cutaneous calciphylaxis, painful and non-healing nodules, plaques, and ulcers may appear, increasing morbidity for patients. Diagnosis can be difficult, and the condition can clinically appear similar to other dermatological diseases, especially in non-uremic patients. Currently, skin biopsy with histological analysis is the most reliable method to help diagnose the condition. In certain cases, the use of medical imaging may be helpful. Treatment of pain in this condition can be difficult and should be multimodal and include wound care as well as modification of risk factors. Analgesic options include opioids, nonsteroidal anti-inflammatory drugs (NSAIDs), as well as analgesic options that are targeted for specific patients. Here we review how massively parallel, or next-generation, sequencing has changed the field of genetic neuromuscular disorders, and anticipate future benefits of recent technological innovations such as RNA-seq implementation and detection of tandem repeat expansions from short-read sequencing.In early German ecology, the key concept used to refer to a synecological unit was Biozönose (biocoenosis). Taken together with the concept of the Biotop (biotope), it was also understood as an integrated higher-order unit of life, sometimes called a "Holozön" (holocoen). These units were often perceived as having properties similar to those of individual organisms, and they informed the mainstream of German ecology until at least the late 1960s. Here I ask how "organismic" these concepts really were and what conceptual problems they entailed. To do so, I focus on some almost forgotten dissident positions, especially those of (German-born) Friedrich Simon Bodenheimer and Fritz Peus, which I contrast with the mainstream German ecology of the time. In a radical paper published in 1954 that postulated the "dissolution of the concepts of biocoenosis and biotope", Peus in particular elicited a forceful response from many prominent German ecologists. An analysis of the ensuing debate, including especially a colloquium held in 1959 that was partly inspired by Peus' paper, is helpful for sifting the various arguments proffered with respect to a quasi-organismic perception of the biocoenosis in German speaking ecology. Although German mainstream ecologists rejected the notion of the biocoenosis as a superorganism, ontological holism was quite common among them. Additionally, the mainstream concept of the biocoenosis was plagued by several methodological problems and much conceptual confusion, to which the "dissidents" rightly pointed. Some of these problems are still pertinent today, e.g. in connection with more modern concepts such as "ecosystem".Since the first issue of the International Journal of Biometeorology in 1957, a total of 135 papers have reported on research in or of African countries. The majority of these have been on topics of animal biometeorology (36%), and the greatest proportion (24%) are situated in Nigeria. There has been a considerable increase in papers on African biometeorology since 2011, with those from this past decade accounting for 58% of all African papers in the journal. This occurs concurrent to an increase in the total number of papers published in the journal, driven by a move to the Editorial Manager system. While 66% of the papers on African biometeorology in the journal are authored by at least one person with an affiliation in the African continent, only 15 African countries are represented in the total authorship. As much of the African continent is projected to experience climatic changes exceeding the global mean, as much of the region is involved in animal and plant farming, and as seasonally-fluctuating and climatically affected diseases are common place, this low representation of work in Africa is surprising.
