The electrochemical activity of catalysts strongly depends on the uniform distribution of monodisperse Pt nanoparticles without aggregates. Here, we propose a new hydrolysis-assisted smearing method for Pt loading on a free-standing paper-type electrode. Polyacrylonitrile (PAN)-based nanofiber paper was used as the electrode, and it acted as a Pt support. Hydrolysis of the electrode tripled the number of active nucleation sites for Pt adsorption on the PAN nanofibers, thereby significantly enhancing the wettability of the nanofibers. This facilitated the uniform distribution of Pt nanoparticles without aggregate formation up to 40 wt% (about 0.8 mg/cm2) with a particle size of about 3 nm. The catalytic current of the hydrolyzed Pt electrode in CH3OH/H2SO4 solution exceeded 213 mA/cm2 Pt mg, which was considerably greater than the current was 148 mA/cm2 Pt mg for an unhydrolyzed electrode.Liebau pump is a tubular, non-peristaltic, pulsatile pump capable of creating unidirectional flow in the absence of valves. It requires asymmetrical positioning of the pincher relative to the attachment sites of its elastic segment to the rest of the circuit. Biological feasibility of such valveless pumps remains a hotly debated topic. To test the feasibility of the Liebau-based pumping in vessels with biologically relevant properties we quantified the output of Liebau pumps with their compliant segments made of a silicone rubber that mimicked the Young modulus of soft tissues. The lengths, the inner diameters, thicknesses of the tested compliant segments ranged from 1 to 5 cm, 3 to 8 mm and 0.3 to 1 mm, respectively. The compliant segment of the setup was compressed at 0.5-2.5 Hz frequencies using a 3.5-mm-wide rectangular piston. A nearest-neighbor tracking algorithm was used to track movements of 0.5-mm carbon particles within the system. The viscosity of the aqueous solution was varied by increased percentage of glycerin. Measurements yielded quantitative relationships between viscosity, frequency of compression and the net flowrate. The use of the Liebau principle of valveless pumping in conjunction with physiologically sized vessel and contraction frequencies yields flowrates comparable to peristaltic pumps of the same dimensions. We conclude that the data confirm physiological feasibility of Liebau-based pumping and warrant further testing of its mechanism using excised biological conduits or tissue engineered components. Such biomimetic pumps can serve as energy-efficient flow generators in microdevices or to study the function of embryonic heart during its normal development or in diseased states.In many industrial sectors continuous processing is already the golden standard to maximize productivity. However, when working with living cells, subpopulation formation causes instabilities in long-term cultivations. In cascaded continuous cultivation, biomass formation and recombinant protein expression can be spatially separated. This cultivation mode was found to facilitate stable protein expression using microbial hosts, however mechanistic knowledge of this cultivation strategy is scarce. In this contribution we present a method workflow to reduce workload and accelerate the establishment of stable continuous processes with E. coli BL21(DE3) exclusively based on bioengineering methods.Christensenellaceae is a family of subdominant commensal bacteria found in humans. It is thought to play an important role in gut health by maintaining microbial symbiosis. Indeed, these bacteria occur at significantly lower levels or are absent in individuals suffering from inflammatory bowel diseases (IBDs). Here, we explored if type species Christensenella minuta (strain DSM 22607) could have the potential to help treat IBDs. We assessed key properties displayed by the bacterium using a combination of in vitro and in vivo assays. We found that while C. minuta is a strict anaerobe, it is also oxygen tolerant. Additionally, we observed that the species produces high levels of acetate and moderate levels of butyrate. We performed deep phenotyping using Biolog microarrays. Using human intestinal cell lines, we discovered that C. minuta demonstrated strong anti-inflammatory activity, resulting in reduced levels of proinflammatory IL-8 cytokines via the inhibition of the NF-κB signaling pathway. Furthermore, C. minuta protected intestinal epithelial integrity in vitro. Finally, in two distinct animal models of acute colitis, C. minuta prevented intestinal damage, reduced colonic inflammation, and promoted mucosal healing. Together, these results indicate that C. check details minuta has potent immunomodulatory properties, underscoring its potential use in innovative microbiome-based IBD biotherapies.Methane emissions along the natural gas supply chain are critical for the climate benefit achievable by fuel switching from coal to natural gas in the electric power sector. For Germany, one of the world's largest primary energy consumers, with a coal and natural gas share in the power sector of 35% and 13%, respectively, we conducted fleet-conversion modelling for reference year 2018, taking domestic and export country specific greenhouse gas (GHG)-emissions in the natural gas and coal supply chains into account. Methane leakage rates below 4.9% (GWP20; immediate 4.1%) in the natural gas supply chain lead to overall reduction of CO2-equivalent GHG-emissions by fuel switching. Supply chain methane emissions vary significantly for the import countries Russia, Norway and The Netherlands, yet for Germany's combined natural gas mix lie with  less then less then  1% far below specific break-even leakage rates. Supply chain emission scenarios demonstrate that a complete shift to natural gas would emit 30-55% (GWP20 and GWP100, respectively) less CO2-equivalent GHG than from the coal mix. However, further abating methane emissions in the petroleum sector should remain a prime effort, when considering natural gas as bridge fuel on the path to achieve the Paris climate goals.The presence of deoxygenated hemoglobin (Hb) results in a drop in T2 and T2* in magnetic resonance imaging (MRI), known as the blood oxygenation level-dependent (BOLD-)effect. The purpose of this study was to investigate if deoxygenated myoglobin (Mb) exerts a BOLD-like effect. Equine Met-Mb powder was dissolved and converted to oxygenated Mb. T1, T2, T2*-maps and BOLD-bSSFP images at 3Tesla were used to scan 22 Mb samples and 12 Hb samples at room air, deoxygenation, reoxygenation and after chemical reduction. In Mb, T2 and T2* mapping showed a significant decrease after deoxygenation (- 25% and - 12%, p  less then  0.01), increase after subsequent reoxygenation (+ 17% and 0% vs. room air, p  less then  0.01), and finally a decrease in T2 after chemical reduction (- 28%, p  less then  0.01). An opposite trend was observed with T1 for each stage, while chemical reduction reduced BOLD-bSSFP signal (- 3%, p  less then  0.01). Similar deflections were seen at oxygenation changes in Hb. The T1 changes suggests that the oxygen content has been changed in the specimen.
