The particle size (PS) and encapsulation efficiency (EE%) of drug-loaded nanoparticles (NPs) may inhibit their cellular uptake and lead to possible leakage of the drug into the systemic circulation at the tumor site. In this work, ultra-high paclitaxel-loaded poly(lactide-co-glycolide) NPs (PTX-PLGA-NPs) with ultra-small sizes were prepared and optimized by adopting the principles of quality by design (QbD) approach. The optimized PTX-PLGA-NPs showed ultra-small spherical particles of about 53 nm with EE% exceeding 90%, a relatively low polydispersity index (PDI) of 0.221, an effective surface charge of -10.1 mV, and a 10-fold increase in the in vitro drug release over 72 h relative to free drug. The cellular viability of pharynx carcinoma cells decreased by almost 50% in 24 h following treatment with optimized PTX-PLGA-NPs, compared to only 20% from the free drug. The intracellular uptake of PTX-PLGA-NPs was highly favored, and the antitumor activity of PTX was remarkably improved with a reduction in its half maximal inhibitory concentration (IC50), by almost 50% relative to free drug solution. These results suggest that the optimal critical formulation parameters, guided by QbD principles, could produce PLGA-NPs with remarkably high EE% and ultra-small PS, resulting in enhanced cellular uptake and efficacy of PTX.International outbreaks of listerial infections have become more frequent in recent years. Listeria monocytogenes, which usually contaminates food, can cause potentially fatal infections. Listerial cerebritis is a rare disease that is encountered mostly in immunocompromised or elderly patients. Saracatinib However, listerial brainstem encephalitis (mesenrhombencephalitis or rhombencephalitis) is found in persons who were formerly in good health, and recognizing this disease, particularly at its early stages, is challenging. Listerial brainstem encephalitis has high mortality, and serious sequelae are frequently reported in survivors. Early recognition and correct diagnosis, as well as the timely use of appropriate antibiotics, can reduce the severity of listerial infections. The trigeminal nerve is proposed as a pathway through which L. monocytogenes reaches the brainstem after entering damaged oropharyngeal mucosa or periodontal tissues. This review introduces the clinical manifestations, pathology, magnetic resonance imaging (MRI) findings, diagnosis, and treatment of listerial brainstem encephalitis. Moreover, it proposes that L. monocytogenes may also invade the brainstem along the vagus nerve after it infects enteric neurons in the walls of the gastrointestinal tract.Polyoxometalates (POMs) has proved its advantage in constructing high-performance nanocomposite membranes such as catalytic membranes, adsorptive membranes, and forward osmosis membranes. However, it is challenging or tedious to characterize its distribution and effect on the membrane structures due to the equipment resolution limitation, discrete nano-scaled structures of POMs, and limited doping amount compared to the polymeric membrane matrix. In this paper, POMs-functionalized polyvinylidene fluoride (PVDF) membranes were fabricated by phase inversion combined with the sol-gel method, and electrochemical impedance spectroscopy (EIS) was utilized to analyze the nanocomposite membrane intrinsic properties. Through adjusting the additives in the sol-forming process, a set of membranes with varied intrinsic properties were developed accordingly. The wetting degree of the membranes related to the hydrophilic nature of the membrane surfaces had a crucial influence on the impedance measurements at the early stage. Through EIS analysis, it was demonstrated that the amination of the membrane matrix through (3-aminopropyl)trimethoxysilane (APTMS) treatment and the immobilization of POMs through electrostatic attraction would not generate new pore structures into the membrane and only alter the membrane surface roughness and composition. To my knowledge, it is the first time that EIS was utilized to characterize the hydrophilicity of the membranes and pore structures of the POMs-modified membranes. Our findings indicate that EIS can provide valuable information for probing the structures of other nano-materials-incorporated membranes. In people with haemophilia (PWH), elastic band training is considered an optimal option, even though the literature is scarce. The aim was to evaluate normalized electromyographic amplitude (nEMG), rate of perceived exertion (RPE), pain, tolerability, and possible adverse effects during the knee extension exercise using multiple elastic resistance intensities in PWH. During a single session, 14 severe PWH undergoing prophylactic treatment performed knee extensions without resistance and with different intensity levels of elastic resistance. nEMG was measured for the knee extensors and participants rated their RPE, tolerability and pain intensity after each condition. Patients had to report the possible adverse effects after the session. In most of the cases, an nEMG increase is only evidenced after increasing the resistance by two to three levels. Significant associations were found between RPE and the nEMG (ρ = 0.61), as well as between the elastic resistance level and nEMG (ρ = 0.69) and RPE (ρ = 0.71). All conditions were generally tolerated, without increased pain, and no adverse effects were reported. A wide variety of elastic resistance intensities during the knee extension are safe, tolerated, and do not increase knee pain in the majority of severe PWH undergoing prophylactic treatment. The enhancement of the ultraviolet (UV) photodegradation resistance of biodegradable polymers can improve their application efficacy in a natural environment. In this study, the hexadecylamine modified layered zinc phenylphosphonate (m-PPZn) was used as a UV protection additive for poly(butylene adipate-co-terephthalate) (PBAT) via solution mixing. The results from the Fourier transform infrared spectroscopy (FTIR) and wide-angle X-ray diffraction analysis of the m-PPZn indicated the occurrence of hexadecylamine intercalation. FTIR and gel permeation chromatography were used to characterize the evolution of the PBAT/m-PPZn composites after being artificially irradiated via a light source. The various functional groups produced via photodegradation were analyzed to illustrate the enhanced UV protection ability of m-PPZn in the composite materials. From the appearance, the yellowness index of the PBAT/m-PPZn composite materials was significantly lower than that of the pure PBAT matrix due to photodegradation. These results were confirmed by the molecular weight reduction in PBAT with increasing m-PPZn content, possibly due to the UV photon energy reflection by the m-PPZn.
