Sprint interval training (SIT) has emerged as a time-efficient training regimen for young individuals. Here, we studied whether SIT is effective also in elderly individuals and whether the training response was affected by treatment with the antioxidants vitamin C and E. Recreationally active elderly (mean age 65) men received either vitamin C (1 g/day) and vitamin E (235 mg/day) or placebo. Training consisted of nine SIT sessions (three sessions/week for three weeks of 4-6 repetitions of 30-s all-out cycling sprints) interposed by 4 min rest. Vastus lateralis muscle biopsies were taken before, 1 h after, and 24 h after the first and last SIT sessions. At the end of the three weeks of training, SIT-induced changes in relative mRNA expression of reactive oxygen/nitrogen species (ROS)- and mitochondria-related proteins, inflammatory mediators, and the sarcoplasmic reticulum Ca2+ channel, the ryanodine receptor 1 (RyR1), were blunted in the vitamin treated group. Western blots frequently showed a major (>50%) decrease in the full-length expression of RyR1 24 h after SIT sessions; in the trained state, vitamin treatment seemed to provide protection against this severe RyR1 modification. Power at exhaustion during an incremental cycling test was increased by ~5% at the end of the training period, whereas maximal oxygen uptake remained unchanged; vitamin treatment did not affect these measures. However, our understanding of the immunological basis of this severe complication is still limited. In this study, we developed a flow cytometric approach to precisely characterize the immune cell composition in periprosthetic joints. Using high-dimensional multi-parametric data, we defined, for the first time, the local immune cell populations of artificial joints. We identified significant differences in the cellular distribution between infected and non-infected samples, and revealed that myeloid-derived suppressor cells (MDSCs) act as potential regulators of infiltrating immune cells in PJI. Further, we developed an algorithm to predict septic and aseptic samples with high sensitivity and specificity, that may serve as an indispensable addition to the current criteria of the Musculoskeletal Infection Society. This study describes a novel approach to flow cytometrically analyze the immune cell infiltrate of joint fluid that not only improves our understanding of the pathophysiology of PJI, but also enables the development of a novel screening tool to predict infection status. Our data further suggest that pharmacological targeting of MDSCs represents a novel strategy for addressing PJI.In this study, we aimed to process the biomimetic function surface by designing a prototype for modeling the pits on a dung beetle body and the abdomen of a desert viper, and by using high speed milling and controlling the ratio of row spacing to feed rate. Firstly, we conducted three-dimensional parametric modeling and static analysis of the bionic functional surface using 3D modeling software UGNX (12.0, SIEMENS AG, Munich, Germany) and finite element analysis software ABAQUS (2018, Dassault, Providence, RI, USA). Then, the analysis results were imported into the fatigue life analysis software nCode (2018, HBM United Kingdom Ltd., South Yorkshire, UK) to simulate the fatigue characteristics of different bionic pit morphology models. Per the simulated tensile fatigue testing machine, the result shows that the minimum fatigue life value of the quadrilateral pit surface of the simulated dung beetle is one and four times higher than the hexagonal pit morphology and the irregular pit morphology, respectively, whereas the maximum fatigue damage is lower by one and five orders of magnitude, respectively. The quadrilateral pit surface on the biomimetic dung beetle body has better fatigue resistance, which can considerably improve the fatigue damage distribution state and the fatigue life of hardened steel die surfaces. The influential regulation of milling parameters on fatigue performance was studied and the results show that the fatigue resistance of the model is optimal when milling parameters are row spacing of 0.4 mm, loading space of 0.2 mm, and milling depth of 0.3 mm. The quadrilateral dimensions formed by milling are highly similar to those of a dung beetle body proving that a certain reduction in milling process depth can increase the structural fatigue resistance. However, because the growth and preservation of wood are greatly affected by the environment, it often contains different types of defects that affect its performance and ornamental value. To solve the issues of high labor costs and low efficiency in the detection of wood defects, we used machine vision and deep learning methods in this work. A color charge-coupled device camera was used to collect the surface images of two types of wood from Akagi and Pinus sylvestris trees. A total of 500 images with a size of 200 × 200 pixels containing wood knots, dead knots, and checking defects were obtained. The transfer learning method was used to apply the single-shot multibox detector (SSD), a target detection algorithm and the DenseNet network was introduced to improve the algorithm. The recorded data from the strain profile at the mid-span of the frame girder indicated that there was a considerable membrane effect, which delayed the strain growth of rebars, yielding appreciable bearing capacity. Thus, two original approaches to predicting the ultimate load of this novel structure are proposed, considering limit analysis using the upper-bound method and the membrane effect, with the latter closely linked to the experimental results.
